GelCode Blue Safe Protein Stain
Thermo Scientific GelCode Blue Safe Protein Stain enables quick and safe in-gel protein detection. With sensitivities down to 9 ng per protein band, GelCode Blue Safe Protein Stain delivers high sensitivity at low cost. The stain is Coomassie G-250-based and is non-hazardous, odorless, non-flammable and non-corrosive to skin. In addition, the stain does not require special hazardous shipping as per US Department of Transportaton (DOT) guidelines, minimizing product shipping costs.
To prepare the stain, add the Activator Crystals, shake and the stain is ready. Unlike traditional coomassie stains, GelCode Blue Safe Protein Stain requires no tedious methanol or acetic acid protein fixation before staining, nor does it require special destaining solutions, saving time and minimizing reagent costs. Just wash gel in water, stain with activated Gelcode Blue Safe Protein Stain, and then destain in water to generate crisp blue bands that are easily distinguished from an ultra-low background.

GelCode Blue Safe Stain Protocol
Step 1. Wash the gel three times with deionized water (15 minutes)
Step 2. Add activated GelCode Blue Safe Protein Stain (15 minutes - 1 hour)
Step 3. Destain in ultrapure water (1-2 hours or overnight)

Detection levels of GelCode Blue Safe Protein Stain
A mixture of proteins (myosin, ß-galactosidase, phosphorylase B, BSA, ovalbumin, carbonic anhydrase, soybean trypsin inhibitor, lysozyme, aprotinin) was electrophoresed on 4-20% Precise Protein Gels (Product # 25224). The gels were stained with GelCode Blue Safe Protein Stain for five minutes, 15 minutes, and one hour and destained for one hour in ultrapure water. Lane 1: 1,000 ng, Lane 2: 500 ng, Lane 3: 250 ng, Lane 4: 125 ng, Lane 5: 63 ng, Lane 6: 31 ng, Lane 7: 16 ng, Lane 8: 8 ng
Quantitative densitometry performance of protein stains
Varying concentrations of BSA (2 ng to 1,000 ng) were electrophoresed on 4-20% Precise Protein Gels (Product #25224). Gels were washed in water (3 x 5 min) and stained with Thermo Scientific Gelcode Blue Safe Protein Stain or stains from other suppliers for one hour and destained with water for 1 hour. Densitometry values were obtained using Kodak MI software.
