Recombinant Protein Aおよびその標識複合体
● 天然型プロテインA(Natural Protein A)
天然型プロテインAは黄色ブドウ球菌(S.aureus)細胞壁由来の1本鎖のポリペプチドで、数種のIgG抗体のFc部位を介して分子内の4つの結合サイトで特異的に結合します。熱に耐性があり、4M尿素や6Mグアニジン塩酸塩に曝された後でもネイティブな立体構造を維持することが出来ます。IgGに対するアフィニティはIgGの種やサブクラスによる差が大きく、またpH依存的です。ヒトでは、IgG1、IgG2、IgG4は強く結合しますが、IgG3では結合が確認されていません。マウスではTris-HClやPBSのような標準的なバッファー条件ではIgG2a、IgG2b、IgG3は強く結合しますが、IgG1は弱い結合のみが確認されています。ラットIgGでは、サブクラスによらず殆ど結合しません。この性質を利用して、血清からのIgGサブクラスの精製（アフィニティの弱いサブクラスを通過・回収）も行われています。ただし、アルブミンのような血清タンパク質やIgM・IgD・IgAなどのIgクラスも通過画分に存在します。
● 組換え型プロテインA(Recombinant Protein A)
非病原性の桿菌に発現させた組替え型で、遺伝子的に結合に不必要な部位を除去したIgG結合サイトを5つ含む、44.6KDaのタンパク質です。(ブドウ球菌が産生する)エンテロトキシンフリーを厳しく要求する系では組替え型を使うべきと考える研究者もいるようです。
