ExcelArray Human MMP/TIMP Array Kit

Protein microarray slides for multiplex fluorescent ELISA. Simultaneously assay for matrix metalloproteinase markers MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-12, MMP-13, TIMP-1, TIMP-2, TIMP-4.

Thermo Scientific ExcelArray Antibody Sandwich Microarrays are fluorescent ELISA kits for simple, quantitative and simultaneous measurement of up to 12 proteins in 10 samples using an appropriate slide reader. The ExcelArray Human MMP/TIMP Array Kit measures MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-12, MMP-13, TIMP-1, TIMP-2, TIMP-4.

The ExcelArray Platform consists of a glass microscope slide containing 16 wells (subarrays), each of which is printed with triplicate spots of specific antibodies and control elements. The 16-well slide enables accurate multplex analysis of 10 samples against a 6-point standard curve. The kits contain ready-to-use mixtures of standards and biotin-labeled detection antibodies, making the multiplex immunoassay as easy to perform as any traditional single-target ELISA. Secondary detection is accomplished with Streptavidin DyLight 649 Reagent, a probe with red fluorescence that is compatible with conventional fluorescent slide readers

The matrix metalloproteinases (MMPs) are a family of zinc proteases involved in the breakdown of extracellular matrix (ECM) in normal physiological processes, such as embryonic development, tissue and bone remodeling, wound healing, and angiogenesis. Deregulation of MMPs has been implicated in numerous diseases, including arthritis, cardiovascular disorders, and cancerIn general, MMPs share four common structural motifs: a pro-peptide domain, a catalytic domain (including the coordinated Zn++ ion at the active site), a hinge region, and a hemopexin-like domain. MMPs are synthesized and secreted as inactive pro-enzymes that subsequently become proteolytically cleaved and activated. This proteolytic activation can be mediated by serine proteases or other MMPs. MMP activity is also regulated at the level of transcription, by NO-mediated S-nitrosylation, and by the endogenous MMP inhibitors, tissue inhibitors of metalloproteinases (TIMPs). MMPs can be upregulated in response to a wide range of stimuli, including adhesion molecules, growth factors, cytokines, and hormones.
