Title: Does Mass Spectrometry Play a Role in Structural Biology?
Many important problems in biology cannot be solved by the high resolution methods of X-ray crystallography and NMR, and they are well beyond the low resolution methods of CD, fluorescence, absorbance, Raman, and calorimetry. Our hypothesis is that mass spectrometry coupled with chemical footprinting can provide a medium-resolution method to solve problems that involve large protein complexes, protein-protein interaction interfaces, binding sites, affinities with ligands and proteins, and properties in complex media where solid state structure may not be relevant. To provide strategies to solve these problems, we are adapting H/D amide exchange and developing new approaches to fast covalent footprinting with reactive radicals and other species (we call the latter method FPOP of fast photochemical oxidation of proteins). We will demonstrate that these approaches can follow protein folding, define binding interfaces, determine binding orders, measure affinites, and uncover allosteric interactions. All of this relies on the multitude of analytical proteomic methods and the advances in separation and mass spectrometric instrumentation.

Title: ORBITRAP MASS SPECTROMETRY : RECENT DEVELOPMENTS
The talk describes new technology advances implemented in recent extensions of two major families of Orbitrap instruments, LTQ Orbitrap and Exactive Plus. These advances are associated with the increase of spectral acquisition rate, sensitivity and resolving power and open new possibilities in accurate mass analysis, especially of proteins and complex mixtures.
